Description
The Synuclein-alpha (Phospho-Ser129) Cell-Based ELISA Kit is a convenient, lysate-free, high throughput and sensitive assay kit that can monitor Synuclein-alpha (Phospho-Ser129) protein expression profile in cells. The kit can be used for measuring the relative amounts of Synuclein-alpha (Phospho-Ser129) in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on Synuclein-alpha (Phospho-Ser129).

- Microplate reader able to measure absorbance at 450 nm and/or 595 nm for Crystal Violet Cell Staining (Optional)
- Micropipettes with capability of measuring volumes ranging from 1 ul to 1 ml
- 37% formaldehyde (Sigma Cat# F-8775) or formaldehyde from other sources
- Deionized or sterile water
- Squirt bottle, manifold dispenser, multichannel pipette reservoir or automated microplate washer
- Graph paper or computer software capable of generating or displaying logarithmic functions
- Absorbent papers or vacuum aspirator
- Test tubes or microfuge tubes capable of storing 1 ml

We are interested in buying the Synuclein-alpha (Phospho-Ser129) Colorimetric Cell-Based ELISA Kit. If it is guaranteed that the antibody recognizes specifically human synuclein but not rat synuclein.

Answer
We conducted a sequence comparison between the human and rat alpha synuclein proteins and there is a high sequence homology. Out of the 140 amino acids, 133 amino acids are both shared between human and rat. Which infers that, it is unlikely to have any antibody you will find to alpha-synuclein will definitely not cross-react with rat because there is no sequence within this protein that varies significantly between rat and human. Our product EKC2126 has been tested with rat, mouse and human and showed positive result in mouse and human. But this will not conclude that it will not work in rat since with some optimization, the rat protein could also be detected.

Could you tell me if this product EKC2126 recognize rat samples or not? Although it is written in the datasheets that does not react with rat. If ever, what rat sample did you test?

Results shows negative in rat during our initial testing and this Synuclein-alpha (Phospho-Ser129) Colorimetric Cell-Based ELISA Kit, we cannot recommended for use in rat. Yet, based on our sequence analysis, it shares 100% identity with rat. For additional information, we previously tested rat brain, heart, kidney and spleen tissues lysates.

For EKC2126 Synuclein-alpha (Phospho-Ser129) Colorimetric Cell-Based ELISA Kit shows positive on the following reactions; Peptide-Coated ELISCoat immunogen phospho-peptide and non-phospho-peptide (pair) in 96-well plate. Reading ratio (phospho:non-phospho) is higher than 5:1. Cell-Based ELISA; Grow cells in 96-well plate. Fix cells. Detect with this antibody. Antibody-Microarray testing; Array antibodies on a slice. Label cell lysate with Cye3. Hybridize Cye3-labelled cell lysate with the slice. Scan the slice after washing. This process detects native proteins. Additionally, we have not tested sandwich ELISA, so we do not have any information about antibody pairs.
